DM1-SMe is a small molecule that has been shown to have anticancer activity. It binds to the HER2 receptor on the surface of cancer cells and blocks cell growth, leading to tumor cell death. DM1-SMe has been shown to enhance the effectiveness of other drugs such as maytansine in animal studies. DM1-SMe is a conjugate of DM1 and SMe, which are chemical derivatives of maytansinol. The SME group is attached to the maytansinol structure at C21, which can be cleaved off by cellular esterases in order to release DM1. This conjugate has been found to inhibit human rhabdomyosarcoma cells in vitro and in vivo.
Molecules with similar structures have also been found to inhibit tumor xenografts in mice when used with other chemotherapeutic agents such as cyclophosphamide or doxorubicin.
