Descrizione:A peptide derived from the Hrk (Harakiri) protein, which is a pro-apoptotic member of the BH3-only family within the larger Bcl-2 family of proteins. The BH3 domain of Hrk, contained in the Hrk BH3 peptide, is the critical region responsible for promoting apoptosis by interacting with and neutralizing anti-apoptotic proteins like Bcl-2, Bcl-xL, and Mcl-1.The BH3 domain within the Hrk BH3 peptide binds to anti-apoptotic proteins, disrupting their ability to prevent apoptosis. This releases pro-apoptotic proteins like Bax and Bak, allowing them to oligomerize and permeabilize the mitochondrial outer membrane. This leads to the release of cytochrome c and the activation of downstream caspases, which execute cell death.Hrk specifically targets Bcl-xL and Mcl-1 more efficiently than some other BH3-only proteins, making it a potent inducer of apoptosis in cells where these anti-apoptotic proteins are overexpressed, such as in certain cancer cells.
