Suvorexant is a drug that belongs to the class of benzodiazepine derivatives. It is the first-in-class sleep-promoting drug, which has been shown to be effective in the treatment of insomnia. Suvorexant binds with high affinity and selectivity to orexin receptors, which are found in high concentrations in the hypothalamus and are thought to regulate arousal and sleep. This binding leads to an increase in GABA levels, thereby suppressing neural activity and promoting sleep. Suvorexant also has a chiral center, so it can exist as a racemic mixture or as individual enantiomers. The racemic form is prepared by mixing equal amounts of the two enantiomers followed by crystallization from water or ethanol. The hydrochloride salt form is prepared by reacting suvorexant with hydrochloric acid followed by crystallization from water or ethanol.
