GSK-356278 is a phosphodiesterase inhibitor that has been shown to improve the symptoms of Alzheimer's disease. This drug is being developed for the treatment of other conditions, such as depression and schizophrenia. GSK-356278 inhibits PDE5, which is involved in the regulation of neurotransmitter release. This inhibition leads to increased levels of cAMP and subsequent activation of protein kinase A (PKA). PKA is a regulatory enzyme that phosphorylates serine residues on many proteins, including ion channels, receptors, enzymes, and transcription factors. GSK-356278 has also been shown to have an effect on synaptic dysfunction by increasing levels of nucleotides in rat brain tissue.
