Semustina, também conhecido como metil-CCNU, é um agente alquilante sintético utilizado principalmente no tratamento de certos tipos de câncer, particularmente tumores cerebrais e linfomas. Pertence à classe das nitrosoureas, que são conhecidas por sua capacidade de atravessar a barreira hematoencefálica, tornando-as eficazes para malignidades do sistema nervoso central. Semustina funciona introduzindo grupos alquilo no DNA, levando à formação de ligações cruzadas e à subsequente interrupção da replicação e transcrição do DNA, induzindo, em última instância, a morte celular. O composto é tipicamente administrado por via oral e tem uma meia-vida relativamente longa, permitindo efeitos terapêuticos sustentados. Os efeitos colaterais comuns associados ao Semustina incluem mielossupressão, distúrbios gastrointestinais e potencial hepatotoxicidade. Devido ao seu mecanismo de ação, é frequentemente utilizado em combinação com outros agentes quimioterápicos para aumentar a eficácia.
