It binds to the ATP site of the enzyme and prevents the phosphorylation of tyrosine residues on cellular proteins, thereby inhibiting protein synthesis. 2-Keto crizotinib has been shown to inhibit receptors that are involved in angiogenesis and metastasis. This agent is also a competitive antagonist at the epidermal growth factor receptor (EGFR) and blocks activation by binding to its extracellular domain. 2-Keto crizotinib also has immunosuppressive effects, which may be due to its ability to block antibody production.
