CAS:Di-tert-butylether (DTBE) is a solvent that is used in analytical methods to extract organic compounds. DTBE has been shown to inhibit the activities of various enzymes, such as polymerase chain reaction (PCR), which are important for DNA replication, and other enzymes involved in protein synthesis. DTBE also inhibits the activity of human serum albumin by binding with the hydroxyl groups on the protein. It is thought that the enzyme inhibitor binds to a specific site on the enzyme and blocks access to this site by substrates or cofactors, thereby preventing catalysis. DTBE has been shown to have toxic effects on wild-type strains of Escherichia coli, but not on mutant strains. This suggests that it may be possible to develop drugs that are resistant to DTBE toxicity by making changes at a single locus in the bacterial genome.
