VU0071063 is metabolized in the liver, but penetrates the blood-brain barrier and reaches high concentrations in the pancreas. The compound has a low toxicity profile and shows no appreciable cardiotoxicity. VU0071063 inhibits both subunits of ATP synthase, preventing movement of potassium ions across the mitochondrial membrane and increasing intracellular calcium ion levels. This leads to impaired insulin secretion by pancreatic β-cells, as well as decreased ATP production by other cells.
