The amide bond is formed by reaction with ammonia or an amine. Finally, it can be reacted with hydrochloric acid to form the corresponding hydrochloride salt, which is insoluble in water. Impurities such as thionyl chloride and chloroformate can be removed by washing with methylene chloride or chloroform, respectively. Fmoc-Ser(tBu)-OH has been shown to be selective for the formation of hydrophobic bonds between amino acids and is not reactive toward other side chains on the protein.
