Valinomycin is a drug that acts as an inhibitor of the mitochondrial membrane potential and can be used as a model system for studying mitochondrial functions. Valinomycin binds to the cytosolic Ca2+ ion channel and prevents its opening, preventing the release of Ca2+ ions from the cell. This leads to an increase in the mitochondrial membrane potential, which is accompanied by an increase in ATP synthesis. Valinomycin also inhibits drug transporter proteins, including P-glycoprotein, thus making it useful for treating cancer. Valinomycin has been shown to have several physiological effects such as increasing energy metabolism and inhibiting transfer reactions, which are involved in various metabolic pathways.
