Cuspidiol is a natural product that has been identified as a novel inhibitor of the enzyme phosphatase 1B (PTP1B). It is a potent and selective inhibitor of PTP1B, with an IC50 value of 0.27 μM. Cuspidiol may function by binding to the active site of the PTP1B enzyme, blocking access to its ATP substrate, and inhibiting its activity. Cuspidiol has also been shown to inhibit transcriptional activation of genes in human cells by suppressing the expression of c-Fos protein. This inhibition is specific for PTP1B and does not affect other phosphatases. The mechanism by which cuspidiol inhibits PTP1B is currently unknown but may involve binding at the active site or interference with substrate binding.
