It is a cofactor for pyruvate dehydrogenase, alpha-ketoglutarate dehydrogenase, and transketolase. Thiamine triphosphate is required for the production of acetyl-CoA from carbohydrates and amino acids. This coenzyme also participates in the synthesis of thiamin, which is necessary for maintaining normal nerve function. The x-ray crystal structures of thiamine triphosphate with its substrate molecule have been determined by X-ray crystallography. These structures are available at PDB ID 1TMP and 1THP. The reaction solution used to determine these structures was phosphate buffer, pH 7.0, at 30 degrees Celsius.
