Methyl 4-cyano-2-methylbenzoate (Xl888) is a small molecule that is currently under development as an inhibitor of Hsp90. It has been shown to inhibit tumor growth in mouse xenograft models, and to induce tumor regression in preclinical models of human cancers. The mechanism of action for Xl888 is not completely understood, but it may be due to inhibition of the ubiquitin-proteasome pathway by Hsp90 inhibition. Structural studies suggest that Xl888 may also inhibit the activity of other proteins that are involved in cancer progression.
