MMV008138 is an indole alkaloid that is structurally similar to the natural product quinine, which has been used as an antimalarial drug for many years. MMV008138 has been shown to be active against protozoan parasites in nanomolar concentrations and this effect can be blocked by the addition of a competitive inhibitor of its target enzyme, Dihydroorotate Dehydrogenase (DHODH). MMV008138 inhibits DHODH and blocks the production of pyrimidine nucleotides, which are essential for DNA synthesis. It also inhibits the biosynthesis of the viral RNA genome and therefore may have potential use in the treatment of viral infections.
