2-Amino-5-tert-butyl-1,3,4-thiadiazole is a small molecule that inhibits P38 kinase and has shown to be effective in the treatment of osteoporosis. It is also effective against arthritis and other diseases that are related to cytokine activity. 2-Amino-5-tert-butyl-1,3,4-thiadiazole binds to the ATP binding site on P38 kinase and prevents the phosphorylation of its downstream targets. This inhibits the production of proinflammatory cytokines such as IL6 and IL8. This compound does not inhibit JNK or ERK kinases and has low toxicity for human cells. It is currently being used in combinatorial chemistry studies with other compounds to find new treatments for inflammatory diseases such as psoriasis and cancer.
