2-Amino-4-methoxynicotinonitrile (4-AMA) is a novel agent that has been shown to induce DNA damage and cell death in vitro and in vivo. 4-AMA binds to the pyrimidine nucleosides, which are important for DNA synthesis, preventing the incorporation of these nucleosides into DNA. 4-AMA can be administered orally and is able to cross the blood-brain barrier, which may be due to its lipophilic properties. The frequency of tumors in mice was reduced by more than 80% when 4-AMA was administered orally over a period of 2 weeks. Administration of 4-AMA also resulted in significant cytotoxicity in human prostate cancer cells as well as human fibroblasts. This drug also showed a high cytotoxicity against mouse melanoma cells and human fibroblasts exposed to urine samples from patients with bladder cancer.
