5-Acetylsalicylic acid methyl ester is an organic compound that is a reactive acetylation intermediate. It is used as a building block in the synthesis of peptidomimetics. 5-Acetylsalicylic acid methyl ester has been shown to be an inhibitor of kinesin, a motor protein that transports cellular cargo along microtubules. This inhibition was found to occur through the alkylation of key amino acid residues and an induced conformational change in the protein, which blocks its ability to bind to microtubules. Magnesium acts as a cocatalyst, forming magnesium alkoxy radicals with the 5-acetylsalicylic acid methyl ester, which subsequently react with oxygen or other nucleophiles present in solution or inside cells. The reaction mechanism for this process has been modeled using quantum mechanical calculations and molecular dynamics simulations.
