Clofazimine has been shown to be effective against leprosy and tuberculoid leprosy. It binds to the bacterial enzyme β-glucuronidase, which hydrolyzes glucuronides in the lysosomes. This leads to accumulation of glucuronide metabolites and inhibition of protein synthesis. Clofazimine also has anti-inflammatory properties, which may be due to inhibition of prostaglandin synthesis.
