2-Chloro-1-(3-fluorobenzyloxy)-4-nitrobenzene (2FABN) is a reagent that is used in organic synthesis to cleave bonds. It has been shown to be selective for electron-deficient aromatic rings, such as benzene and nitro compounds. 2FABN has high reactivity towards electron-rich aromatic rings, such as phenols and indoles, which limits its use. 2FABN also reacts with water and other impurities present in the reaction solution. Magnetic stirring is required for the reaction to proceed at an optimal rate. 2FABN can be recycled by distilling off the solvent and leaving behind a solid residue. Lapatinib is an example of a drug that contains 2FABN as an impurity. When it was developed, lapatinib was systematically screened for nitro impurities through mass spectrometry analysis of intact drug molecules.
