3-Cyanochromone is an inhibitor of acetylcholinesterase (AChE) that belongs to the class of reversible covalent inhibitors. It has been shown to have a reversible inhibition constant of 2.5 to 4.5 μM for AChE in human serum and amine-binding sites in the active site of AChE. 3-Cyanochromone binds reversibly to the carbonyl group in the active methylene region, which is located near the amino acid tryptophan, thereby inhibiting the enzyme's activity by preventing its reaction with substrates. 3-Cyanochromone also inhibits butyrylcholinesterase (BChE), which is responsible for breaking down molecules of choline into acetylcholine, a neurotransmitter. This inhibition may lead to accumulation of acetylcholine and subsequent cholinergic crisis.
