Didesmethyl erlotinib hydrochloride salt is a cytotoxic drug used in the treatment of cancer. It is a prodrug that is metabolized to an active form, which inhibits the activity of tyrosine kinases, leading to cell death. Didesmethyl erlotinib hydrochloride salt binds to the enzyme elution and then reacts with oxygen, forming a reactive intermediate. This intermediate reacts with deuterated acetonitrile in LC-MS/MS to produce a deuterated adduct that can be monitored using mass spectrometry. The reaction between didesmethyl erlotinib hydrochloride salt and oxygen leads to the formation of two adducts, one of which has been identified as the product of oxidation at C2 and C3. The other adduct has not yet been identified but could be due to oxidation at C1 and C2 or C1 and C3.
