Diethyl (1,3-Dithian-2-yl)phosphonate is a podophyllotoxin analog that has been shown to have enhanced antitumor activity against murine leukemia and L1210 murine leukemia. It is an ancillary in the asymmetric synthesis of β-amino acid derivatives, benzylidene acetals, and podophyllotoxin analogues. The l1210 murine leukemia cells are sensitive to diethyl (1,3-dithian-2-yl)phosphonate because it can form covalent bonds with DNA by electron donation. These compounds are also found to be cytotoxic against other tumor cells including human breast cancer cells.
