Dihydrofolate reductase is a target enzyme that belongs to the family of dihydropteroate synthases. It catalyzes the conversion of dihydrofolate to folate. The enzyme is also known as DHFR, and is one of the most important enzymes in folate metabolism. It plays an essential role in recycling folates and is used as a target for antifolate drugs. Dihydrofolate reductase has been shown to have drug interactions with some inhibitors, such as sulfasalazine or methotrexate. This enzyme has been studied through model systems and x-ray diffraction data obtained from crystals of c. glabrata, a bacterium that causes leprosy.
