FMK 9a is a potent inhibitor of mammalian autophagy. It inhibits the formation of autophagosomes, which are the structures that enclose cellular contents and break down proteins. FMK 9a induces autophagy by cleaving atypical protein kinase C (PKC) and inhibiting the activation of phosphatidylinositol 3-kinase (PI3K). This drug also attenuates lipolysis and prevents the accumulation of lipidated proteins by inhibiting delipidation. Inhibition of autophagy has been shown to be an effective treatment for pathophysiological conditions such as cancer, diabetes, stroke, glaucoma, Parkinson's disease, ALS, and Alzheimer's disease.
