Ethinyl estradiol 3-acetate is an estrogen that binds to the estrogen receptor. It is a synthetic form of estradiol and its chemical name is ethynyl estradiol 3-acetate (EE3A). EE3A is a prodrug of 17β-estradiol, which is the active form of EE3A. The affinity of EE3A for the estrogen receptor-α ligand binding domain has been shown to be as high as that of 17β-estradiol for the same domain. Structural modifications have been made to increase the stimulatory activity of EE3A, yielding increased levels of alkaline phosphatase in liver cells and increased levels of ligand in some tissues. In addition, these structural modifications have led to an increase in the affinity for the estrogen receptor-α ligand binding domain and improved biological response.
