XLR-11 is an indole derivative synthetic cannabinoid acting as a potent agonist at the cannabinoid receptors CB1 and CB2 (EC50 = 98 nM and 83 nM, respectively)1. Its binding to the CB receptors activates G protein+gated inwardly rectifying K+ channels more strongly than Δ9-THC1. In vitro assays in bacterial cultures did not show genotoxicity upon administration, however, XLR11-induced DNA damage and micronuclei formation were shown to be triggered in human lymphocytic cultures as well as in vitro models of buccal mucosa and lung-derived cell cultures, leading to tumorigenesis concerns2. In vitro, also it suppressed calcium influx in hippocampus rat neurons3. In vivo studies on mice and rat models showed moderate hypothermia and depression of locomotor activity1
