5'-Hydroxy thalidomide is a drug used for the treatment of leprosy and cancer. It has been shown to have immunomodulatory effects in mice with an immunodeficiency. 5'-Hydroxy thalidomide is hydroxylated by cytochrome P450 in the liver to form 5-hydroxy thalidomide, which is a potent inhibitor of tumor necrosis factor-α (TNF-α) production. The effect of 5'-hydroxy thalidomide on TNF-α production may be due to its ability to inhibit proinflammatory cytokine synthesis in human liver cells and human hepatocytes in culture.
5'-Hydroxy thalidomide also inhibits the production of TNF-α by neutrophils, monocytes, and macrophages thereby reducing inflammation.
