2-Iodo-4-nitrophenol is a reagent that can be used to detect tyrosinase activity in cells. The reactivity of this compound with tyrosinase is based on the reaction of the thymidylate moiety with the tyrosinase enzyme, which is a vital component in the production of melanin. This compound has been shown to inhibit cellular antioxidant enzymes and amines, leading to increased cellular damage.
2-Iodo-4-nitrophenol reacts with tyrosinase at pH 5 or higher, and it will not react at pH 4 or lower. The detection sensitivity for 2-iodo-4-nitrophenol can be improved by using electrospray ionization mass spectrometry (ESIMS) or liquid chromatography/mass spectrometry (LCMS).
