2-(Isopropylsulphonyl)aniline (2-ISA) is a potent and selective inhibitor of the mitotic checkpoint kinase Mad2, which is implicated in cancer. 2-ISA has been shown to inhibit the mitotic spindle assembly checkpoint by binding to Mad2 and inhibiting its ability to phosphorylate Mad1 and Cdc20. In vitro experiments have demonstrated that 2-ISA blocks cell cycle progression in ht-29 cells, a colon cancer cell line, by preventing their entry into mitosis. 2-ISA has been shown to be an effective inhibitor of tumor growth in vivo. This agent also inhibits sorafenib, a drug that targets the Raf/MEK/ERK signaling pathway and is used for the treatment of cancer. The analogs of 2-ISA are being developed as potential anticancer agents.
