Lactacystin is a natural compound that belongs to the class of diketopiperazines. It has been shown to induce apoptosis in 3T3-L1 preadipocytes, HL-60 cells and myeloma cell lines. Lactacystin also inhibits mitochondrial membrane potential, leading to the release of cytochrome c from the mitochondria into the cytosol where it triggers an increase in pro-apoptotic protein Bax and a decrease in anti-apoptotic protein Bcl-2. This process leads to caspase activation and DNA fragmentation, resulting in cell death.
