Lysyl-5-fluorotryptophyl-Lysine is an optical probe that can be used to detect DNA damage. It has a fluorescence emission maximum at nmr chemical shift of δ = 6.4 ppm and a binding constant of Kd = 5 x 10 M for the apurinic site, which is the most common type of DNA lesion. Lysyl-5-fluorotryptophyl-Lysine can be detected by incubating samples with this dye and monitoring the fluorescence intensity after thermal denaturation. The dye binds to the damaged site and shifts its absorption spectrum to longer wavelengths when it is excited by UV light. The binding constants are determined from titration experiments, in which one molecule of dye is added to a solution containing many molecules of DNA substrate (apurinic sites). This titration curve is measured as a function of dye concentration, while monitoring the increase in fluorescence intensity due to binding.
