Methyl 3-hydroxythiophene-2-carboxylate is an organic compound consisting of a methyl group attached to a thiophene ring. It is a precursor to the drug amiloride, which is used to treat hypertension and edema. The compound can be formed by the reaction of hydrogen chloride with an alcohol in alkaline conditions. It can also be synthesized from 3-chlorothiophene-2-carboxylic acid and methanol in dilute hydrochloric acid. In addition, it can be prepared by chlorination of methyltetrahydrothiophene-2-carboxylic acid in the presence of copper(II) oxide as a catalyst. The compound has been decarboxylated at 200°C and then reacted with chloroform under refluxing conditions to produce amiloride.
