5-Methylnicotinamide is a nicotinamide derivative that inhibits the subunits of 3-aminobenzamide ribonucleotide (3-AR) deaminase, which is involved in renal proximal tubule function. 5-Methylnicotinamide has been shown to inhibit adenosine diphosphate (ADP) levels and cellular ATP levels in vitro. This agent also has an inhibitory effect on nucleotide synthesis and may be used as a radiation protective agent. 5-Methylnicotinamide was also shown to be effective against resistant mutants in cell culture, including monoclonal antibody resistant mutants. The mechanism of action for 5-methylnicotinamide may involve its ability to stimulate picolinic acid production by binding to the nicotinamide riboside kinase enzyme.
