Methyl 5-chloro-2-hydroxy-3-nitrobenzoate is a precursor to the antiemetic drug azasetron. It is synthesized by refluxing methyl 5-chloro-2-hydroxybenzoate with stannous chloride and hydrochloric acid. The reaction solution is evaporated and the product is purified by crystallization. The synthesis of azasetron hydrochloride starts with the amination of ethyl alcohol, which is reacted with nitryl chloride in aqueous solution at 0°C. This reaction produces substances that are removed by extraction and then treated with hydrochloric acid, producing methyl 5-chloro-2-hydroxy-3-nitrobenzoate as an oily residue. Methyl 5-chloro-2-hydroxybenzoate can be produced from methyl 3,5 dichlorobenzoate and sodium hydroxide in ethanol.
