3-Methylpyrrole is a metabolite of pyrrole that has been shown to inhibit the bacterial enzyme gyrase. 3-Methylpyrrole inhibits bacterial DNA replication and transcription by binding to the active site of the enzyme and preventing ATP from binding. The functional theory for this inhibition is that the methyl group in 3-methylpyrrole binds to the hydroxyl group in the enzyme, which prevents it from binding with ATP. This leads to a decrease in ATP levels, which causes an increase in AMP levels. The increased AMP levels then cause an increase in ADP levels, which leads to a decrease in energy production and cell death. 3-Methylpyrrole also has antimicrobial activity against Gram-positive bacteria such as Staphylococcus aureus but not Gram-negative bacteria such as Escherichia coli or Pseudomonas aeruginosa.
