2-Methylbutyrylglycine (2MBG) is a metabolite of the amino acid leucine that is excreted in the urine. 2MBG is formed from the metabolism of leucine by cytosolic methylmalonyl-CoA mutase, which converts methylmalonyl-CoA to succinyl-CoA. 2MBG can be used as an indicator for leucine metabolism and for diagnosing metabolic disorders such as the deficiency of enzymes involved in fatty acid oxidation. The presence of 2MBG in urine samples may also be related to an increased risk of developing type II diabetes and cardiovascular disease. The enzyme activities associated with 2MBG are dependent on age, sex, diet, and physical activity levels.
