3-Methyl-D-phenylalanine is a peroxidase substrate that has been used in enzyme assays to measure the activity of peroxidases. It has been shown to inhibit horseradish peroxidase and other peroxidases, which catalyze the conversion of hydrogen peroxide to water and oxygen. 3-Methyl-D-phenylalanine reacts with hydrogen peroxide to produce colorimetric end products. Mutant enzymes that lack a functional heme prosthetic group are readily detected using this substrate. 3-Methyl-D-phenylalanine also reacts with o-dianisidine to produce a red product, which can be measured by absorbance at 540 nm. The acceptor for this reaction is phenazine methosulfate. This substrate is enantiopure because it contains only one chiral center and therefore only one optical isomer.
