5-Nitrouracil is a piperazine derivative that inhibits the growth of cells by binding to detoxification enzymes and blocking their activity. It also inhibits the synthesis of proteins, which are necessary for cell division. 5-Nitrouracil has been shown to be effective against cancerous cells in culture, as well as inhibiting the growth of tumor cells in mice. The low energy levels of this drug make it an attractive candidate for use in radiotherapy. 5-Nitrouracil binds to DNA at a site that is different from other cytotoxic drugs, such as cisplatin and adriamycin, and causes structural changes in the DNA molecule. The binding constants have been determined using x-ray diffraction data and kinetic studies. Hydrogen bonds between 5-nitrouracil and water molecules may be responsible for its inhibitory effect on enzymes that require water molecules for their function.
