5-Nitro-1H-imidazole-4-carboxylic acid is a hydrophobic natural product that can be used in cancer therapy. It has been shown to inhibit the growth of tumor cells and induce apoptosis. This compound is also active against leukemia, ovarian cancer, and breast cancer cell lines. 5NICA inhibits protein synthesis by blocking purine uptake and inhibiting the enzyme ribonucleotide reductase, which converts ribose into deoxyribose nucleotides. This inhibition leads to the depletion of ATP and an increase in reactive oxygen species (ROS) levels, which ultimately leads to apoptosis. 5NICA has been shown to bind strongly to the active site of ribonucleotide reductase with high affinity, thereby preventing substrate binding and catalysis.
