4-Nitrophenyl chloroacetate (4NPCA) is a chemical compound that is used in the synthesis of cis-UCA, an inhibitor of ornithine decarboxylase. 4NPCA is synthesized by adding a solution of 4-nitrophenol and acetyl chloride to a suspension of urea. The reaction proceeds through an intramolecular hydrogen transfer from the nitro group to urea, forming cis-UCA and 4NPCA. The process is accelerated by thermodynamic, kinetic, and carbonic effects. Reaction rates are dependent on the pH of the solution. Protons stabilize the intermediate species that form during the reaction mechanism. The stabilizing effect of protons slows down the rate at which intermediate products are formed. Gels containing erythrocyte proteins can be used to separate cis-UCA from 4NPCA using gel chromatography.
