Nicotine 1'-N-oxide is a major metabolite of nicotine in human urine. It is not only a metabolic product of nicotine but also an active substance that has been shown to bind to the nicotinic acetylcholine receptor at the neuromuscular junction, where it mimics the effects of nicotine. Nicotine 1'-N-oxide has been shown to be an insulin-sensitive agent that stimulates glucose uptake and utilization in skeletal muscle cells and insulin secretion in pancreatic β-cells. It has also been found to have inhibitory effects on glycogen synthase, which may contribute to its role as a glycogenolytic agent.
