1,7-Naphthyridin-4(1H)-one is a cyclized quinoline derivative. It can be synthesized by hydrolysis of the 3-aminoquinoline with ethyl acetoacetate to form an amide and then reacting with acetylacetone or acetoacetate to produce the desired product. The yield of this process is high, due to the selectivity of the amide to react with a specific carbonyl group on the aromatic ring. The reaction proceeds in three steps: (i) decarboxylation, (ii) addition of an amine to the carboxylic acid moiety, and (iii) formation of a five-membered ring.
The 1,7-naphthyridin-4(1H)-one has been shown to be a potent inhibitor of bacterial DNA gyrase that is active against multidrug resistant bacteria such as Acinetobacter
