Nocistatin is an amide with a potent antinociceptive effect. Nocistatin has been shown to reduce the severity of bowel disease in rats and its effects are similar to those seen with morphine. The high values of nocistatin have been detected in the hippocampus, trigeminal nerve, and spinal cord, which may contribute to its antinociceptive properties. Nocistatin also has a locomotor activity-enhancing effect in mice. Preparative hplc has revealed that nocistatin is a mixture of two compounds: nociceptin and congestive heart peptide (CHP). Nocistatin binds to the ryanodine receptor at low concentrations and activates it at higher concentrations. It also binds to opioid receptors such as kappa-opioid receptors, which may contribute to its analgesic effects.
