Pyrimidine-2-carbothioamide is an inhibitor of the enzyme aromatase, which converts androgens to estrogens. It has been shown to inhibit the biosynthesis of estrogen, and therefore may be used in clinical applications as a potential anticancer drug. Pyrimidine-2-carbothioamide has been shown to inhibit the activity of aromatase by binding to its active site, which is located at its N-terminal region. This binding prevents the formation of an enzyme/substrate complex with the substrate and cofactor that are required for the reaction. The inhibition of this reaction leads to decreased production of estrogen. It has also been shown that pyrimidine-2-carbothioamide can bind to a number of functional groups such as thiols, amines, and hydroxyls.
