1-Z-3-Piperidone is a potent and selective agonist of the calcium-sensing receptor (CaSR). It has been shown to increase parathyroid hormone release in parathyroid gland cells in vitro. 1-Z-3-Piperidone has been used clinically for the treatment of hyperparathyroidism. In this condition, high levels of calcium ions cause overproduction of parathyroid hormone, leading to bone loss and kidney stones. The clinical use of 1-Z-3-piperidone has been optimized with triflates and vinyl groups to improve its pharmacokinetics.
