Triflumizole is a piperonyl butoxide that is used in the treatment of chronic mycoses. It has been shown to be effective against multidrug efflux pump-mediated resistance in some cases. Triflumizole has also been shown to inhibit the growth of many fungi, including Fusarium proliferatum, which causes canker and dieback in plants. Triflumizole has an affinity for lipophilic substances and thus does not penetrate well into the cell membrane. The hydrophobic effect prevents its penetration into mammalian tissue culture cells, limiting its use as an antimicrobial agent. The mechanism of triflumizole's antifungal activity involves interference with DNA synthesis and protein synthesis in the fungus Fusarium proliferatum. This drug also inhibits matrix metalloproteinases that are involved in bone resorption (osteoclasts).
