n-Tert-butyl-alpha-phenylnitrone (PBN) is a reactive agent that has been shown to protect against ischemia-reperfusion injury, neuronal death, and oxidative injury. In vitro assays have shown that PBN binds to the toll-like receptor 4 (TLR4) of human monocytes and activates the cells. It also increases cytosolic calcium in neurons, which protects against neurotoxic injury. PBN has been shown to increase locomotor activity in mice and to stimulate neurogenesis in vitro. It stimulates the production of neurotrophic factors by HL-60 cells and has an effect on energy metabolism. PBN also exhibits anti-inflammatory effects in vivo through its inhibition of microdialysis probes in the brain, as well as physiological effects such as an increase in blood pressure.
