(S)-4-Hydroxymethyl ambrisentan is a selective endothelin receptor antagonist that has been shown to inhibit cell proliferation in human liver and muscle cells. It also inhibits the metabolism of bosentan, a drug used for the treatment of pulmonary hypertension, through inhibition of cytochrome P450 enzymes. (S)-4-Hydroxymethyl ambrisentan has been shown to be safe and effective as an anti-cancer agent in animal studies. In addition, it has been shown to protect against renal transplantation rejection. The pharmacokinetic properties of (S)-4-hydroxymethyl ambrisentan are similar to those of sildenafil, with a half-life of 2 hours and peak concentration at 3 hours after administration. The drug does not interact with other drugs, but there is potential for interactions with drugs that are substrates for CYP3A4 and CYP2D6 enzymes.
