TAPI 2 is a potent and selective inhibitor of the enzyme, pyruvate dehydrogenase kinase (PDK). PDK regulates the transition from aerobic to anaerobic metabolism. TAPI 2 binds to the active site of PDK and prevents it from phosphorylating and inactivating pyruvate dehydrogenase (PDH), which is essential for ATP production. TAPI 2 also has been shown to inhibit the respiratory chain complexes I, III, and IV. This inhibition has been shown to be reversible by scavenging or binding at a different site on the enzyme. In addition, TAPI 2 inhibits DNA replication by preventing DNA polymerases from interacting with DNA-dependent RNA polymerase. This drug has been shown to have a potent inhibitory effect on tumor cell viability in vitro and in vivo.
