5-Methyl-2-(methylthio)pyrimidin-4(3H)-one is a sulfonamide that causes inhibition of HIV replication. It binds to the active site of the reverse transcriptase, which is an enzyme necessary for DNA synthesis. 5-Methyl-2-(methylthio)pyrimidin-4(3H)-one has been shown to inhibit HIV replication in vitro and to have antiviral effects against HIV-1 in vivo. This drug does not bind well to other nucleic acids and does not affect host cell DNA synthesis. 5-Methyl-2-(methylthio)pyrimidin-4(3H)-one is activated by lithiation, which facilitates its binding to the reverse transcriptase active site, when it is present in concentrations of up to 10 mM.
